ABSTRACT OF THE DISCLOSURE A process is provided for preparing and purifying thymidine phosphorylase of bacterial origin which comprises the steps of extracting the crude enzyme from a culture of Escherichia coli into an aqueous medium and purifying the extract of the crude enzyme by subjecting it to a fractionation procedure which includes, adsorption, elution, column chromato- graphy and dialysis steps; the enzyme is particularly employed in a concentrated solution containing at least 5mg protein/ml, in phosphate buffer containing ammonium sulphate; the process permits a higher level of purity to be readily attained.
