Abstract of the Disclosure A method for purifying recombinant tumor necrosis factor from cultures of microorganisms transformed with vectors that code for the production of tumor necrosis factor is disclosed. A sequence of contacting steps is involved, such that the effluent from the first contacting step can be contacted directly with the contacting material used for the second step and similarly, the effluent from the second contacting step can be contacted directly with the contacting material used for the third contacting step. In a preferred embodiment, tumor necrosis factor-containing fermentation broth can be substantially purified by a single pass through a multi-component chromatography system, i.e., the first, second and third contacting materials are each packed in a column configuration, with TNF-containing broth being passed directly through the sequence of columns. The need for intermediate isolation, concentration, reconstitution, and the like of the tumor necrosis factor-containing fractions is therefore avoided by the practice of the present invention.
