A B S T R A C T Process for the large-scale production of a vaccine against poliomyelitis and the resulting vaccine. For each type of poliomyelitis virus used, there is multiplied a cell strain by means of culturing on microcarriers in suspension and by successive passa- ges into increasing volumes of biogenerators, the last of which holds at least 150 liters, into a suitable nu- tritive medium. At the end of the final passage, the li- quid medium is drawn off and replaced by a new liquid me- dium containing no serum. After inoculation, the virus is allowed to develop in suitable temperature, pH, and par- tial oxygen pressure conditions. The liquid suspension is withdrawn after virus culture. The suspension drawn off is filtered. The filtered suspension is concentrated at least 150 times by means of ultrafiltration. A gel filtra- tion of the concentrated suspension is carried out. The suspension obtained is subjected to ion exchange chromato- graphy. The concentrated suspension obtained is diluted with a serum free medium. The suspension thus diluted and purified is inactivated. The suspension of the respective types used are mixed and the individual dosages are pre- pared.
