ABSTRACT Disclosed is a process for chemically modifying natural- ly occurring proteins to produce biologically active semisynthetic enzymes. The process comprises partially denaturing a protein, then contacting the resulting partially denatured protein with an inhibitor of a model enzyme with the desired biological activ- ity and then cross-linking the protein in situ in the presence of the inhibitor to produce a semisynthetic enzyme having the bio- logical activity of the model enzyme. The thusly prepared semi- synthetic enzyme exhibits different biological activity than the native protein.
