A method is described for the rapid, sensitive and accurate determination of sucrose in biological fluids. A subtstrate is pre-coated with a glucose or fructose polymer (dextran, amylose, levan) and a transglycosidase enzyme (dextransucrase, amylosucrase, levansucrase). When the coated substrate is incubated with biological fluids containing concentrations of sucrose, the transglycosidase enzyme transfers monomers of glucose or fructose from the sucrose to the glucose or fructose polymer. The dimensions of the polymer are increased in proportion to the sucrose concentration of the samples. Newly formed polymer is subsequently quantitated in an immunoassay which employs either a combination of a carbohydrate-binding protein (which may be an antibody) and a conjugate of a secondary antibody and a marker enzyme, or a conjugate of a carbohydrate-binding protein and a marker enzyme. The assay is particularly useful in a non-invasive diagnostic test for gastric damage.
On décrit un procédé de détermination rapide, sensible et précise du saccharose dans des fluides biologiques. Un substrat est préenduit d'un polymère de glucose ou de fructose et d'une enzyme transglycosidase. Une fois le substrat préenduit incubé en présence de concentrations de saccharose contenant des fluides biologiques, l'enzyme transglycosidase transfère au polymère de glucose ou de fructose des monomères de glucose ou de fructose provenant du saccharose. Les quantités du polymère sont accrues par rapport à la teneur des prélèvements en saccharose. Le polymère nouvellement formé est ensuite mesuré dans un dosage immunologique qui met en oeuvre soit une combinaison d'une protéine à liaison glucidique (pouvant être un anticorps) et d'un conjugué d'un anticorps secondaire et d'une enzyme marqueur soit un conjugué d'une protéine à liaison glucidique et d'une enzyme marqueur. Ce dosage est précis pour des concentrations de saccharose inférieures à 1µg/mL. Aucune interférence n'a été observée pour des concentrations de glucose jusqu'à 25 nM. Ce dosage de détection du saccharose est particulièrement efficace pour des tests de diagnostic non invasifs concernant des affections gastriques.
