A method of manufacturing an itraconazole oral dosage from that is substantially free of residual methylene chloride comprises the steps of: (a) providing a working solution comprising an alcohol, a strong acid (preferably an inorganic acid or organic sulphonic acid), itraconazole, a water-soluble polymer, and water, with the itraconazole and the strong acid preferably present in the working solution in a ratio of 1 Mole itraconazole to 1-3 Moles acid; (b) providing particles formed from a pharmaceutically acceptable core material; (c) combining the working solution with the particles to produce itraconazole-coated particles; (d) drying the itraconazole-coated particles; and(e) forming the dried itraconazole-coated particles into an itraconazole oral dosage form that is substantially free of residual methylene chloride. The products of such methods and methods of use thereof are also disclosed.

L'invention concerne un procédé permettant de produire une forme posologique à administration orale d'itraconazole, sensiblement exempte de chlorure de méthylène résiduel. Ce procédé comprend les étapes suivantes : (a) on prend une solution préparée comprenant un alcool, un acide fort (de préférence un acide inorganique ou un acide sulfonique organique), de l'itraconazole, un polymère soluble à l'eau et de l'eau, le rapport itraconazole-acide fort étant de préférence de 1 mole itraconazole pour 1-3 moles d'acide; (b) on prend des particules composées d'un composant de noyau pharmaceutiquement acceptable; (c) on combine la solution préparée avec les particules afin de produire des particules enrobées d'itraconazole; (d) on sèche ces particules enrobées d'itraconazole; et (e) on forme ces particules séchées enrobées d'itraconazole de manière à produire une forme posologique d'itraconazole à administration orale, sensiblement exempte de chlorure de méthylène résiduel.
