A vector comprising a first DNA sequence which is complementary to at least part of an alphavirus RNA genome and having the complement of complete alphavirus DNA genome replication regions, a second DNA sequence encoding a paramyxovirus protein, particularly a respiratory syncytial virus fusion (RSV F) protein or an RSV F protein fragment that generates antibodies that specifically react with RSV F protein, the first and second DNA sequences being under the transcriptional control of a promoter is described. Such vector may be used to produce an RNA transcript which may be used to immunize a host, including a human host, to protect the host aginst disease caused by paramyxovirus, particularly respiratory syncytial virus, by administration to the host. The RNA transcript may be formed by linearization of the vector through cleavage at a unique restriction site in a plasmid vector and then transcribing the linear molecule.
L'invention se rapporte à un vecteur comprenant une première séquence d'ADN, complémentaire à une partie au moins du génome d'un ARN d'alphavirus et possédant le complément des régions complètes de réplication du génome d'un ADN d'alphavirus, une deuxième séquence d'ADN codant pour une protéine de paramyxovirus, en particulier pour une protéine de fusion du virus respiratoire syncytial (SRV F) ou un fragment de la protéine de SRV F qui génère les anticorps réagissant spécifiquement avec la protéine SRV F. Les première et deuxième séquences d'ADN se trouvent sous le contrôle transcriptionnel d'un promoteur. Le vecteur décrit dans l'invention peut être utilisé pour fabriquer un transcript d'ARN pouvant être administré à un hôte (y compris un hôte humain) afin de l'immuniser et de le protéger contre une maladie causée par un paramyxovirus, en particulier par le virus respiratoire syncytial. On peut former le transcript d'ARN en procédant à la linéarisation du vecteur par le clivage sur un site de restriction unique dans un plasmide vecteur et par la transcription ultérieure de la molécule linéaire.
