A composition for transmucosally administering a drug to the oral cavity comprises an adhesive layer comprising a hydrophilic polymer having one surface adapted to contact a first tissue of the oral cavity and adhere thereto when wet and an opposing surface in contact with and adhering to an adjacent drug-containing layer comprising an effective amount of a drug and optionally an effective amount of a permeation enhancer, wherein the drug- containing layer is adapted to contact and be in drug transfer relationship with a mucosal tissue of the oral cavity when the adhesive layer contacts and adheres to the first tissue. Preferred drugs include peptides, such as glucagon-like insulinotropic peptides. A method of transmucosally administering a drug to the oral cavity is also disclosed.
Une composition permettant d'administrer à travers la muqueuse un médicament dans la cavité orale comprend une couche adhésive renfermant un polymère hydrophile dont une surface est prévue pour être en contact avec un premier tissu de la cavité orale et pour adhérer à ce dernier lorsqu'elle est humide, et dont une surface opposée se trouve en contact avec une couche adjacente contenant un médicament sur laquelle elle adhère, cette couche adjacente contenant un médicament comprenant une quantité efficace d'un médicament et facultativement une quantité efficace d'un accélérateur de pénétration; la dite couche contenant le médicament étant adaptée pour se trouver en contact avec un tissu de la muqueuse de la cavité orale et pour assurer le transfert du médicament à travers le tissu de la muqueuse lorsque la couche adhésive se trouve en contact avec le premier tissu auquel elle adhère. Les médicaments préférés comprennent des peptides tels que les peptides insulinotropes du type glucagon. On décrit également un procédé d'administration de médicament dans la cavité orale à travers la muqueuse.
