The present invention relates to materials and methods for the quantitative and qualitative analysis of gene expression during targeted differentiation of isolated homozygous stem cells, and to materials and methods for accumulating sequence tags sampled from a population of expressed genes. More particularly, the present invention relates to a method for constructing a model of cellular development and differentiation comprising the steps of: (1) producing a homogenous population of homozygous stem (HS) cells; (2) directing the differentiation of the HS cells to arrive at desired mature (somatic) cells, group of cells or tissue; (3) periodically sampling HS cells undergoing differentiation to extract total cellular RNA produced at various sampling stages; (4) isolating mRNA from the total cellular RNA; and (5) constructing a cDNA library representative of proteins produced and expressed at various stages of differentiation, over the course of time. The present invention further relates to sequencing and cataloguing isolated mRNAs and cDNAs at various stages of differentiation of HS cells, producing and cataloguing isolated polypeptides sequences encoded by same, and screening the cDNA libraries. I vitro and in vivo cultures of homozygous stem cell are also described; such cultures find utility both as models of differentiation as well as sources of differentiated tissues and organs.

L'invention porte sur des matériaux et procédés servant à l'analyse quantitative et qualitative de l'expression de gènes lors d'une différenciation ciblée de cellules souches homozygotes isolées, et sur des matériaux et procédés servant à accumuler des marqueurs de séquences échantillonnés dans la population de gènes exprimés. L'invention porte plus particulièrement sur une méthode de construction d'un modèle de développement et différenciation cellulaires comportant les étapes suivantes: (1) production d'une population homogène de cellules souches homozygotes (cellules HS); (2) gestion de la différentiation des cellules HS afin d'obtenir les cellules, groupes de cellules, ou tissus, matures (somatiques) voulus; (3) échantillonnage périodique de cellules en cours de différentiation et extraction de la totalité de l'ARN des cellules produites aux différents stades de l'échantillonnage; (4) isolement de l'ARNm de la totalité de l'ARN des cellules; et (5) construction progressive d'une bibliothèque d'ARNc représentative des protéines produites et exprimées aux différents stades de différenciation. L'invention porte en outre: (a) sur le séquençage et le catalogage des ARNm et ADNc isolés aux différents stades de différenciation des cellules HS; (b) sur la production et le catalogage de séquences de polypeptides isolées codées par lesdits ARNm et ADNc; (c) sur le criblage des bibliothèques d'ADNc; et (d) sur des cultures <i>I vitro</i> et <i>in vivo</i> de cellules HS, lesdites cultures étant utiles, et comme modèles de différenciation, et comme sources de tissus et d'organes différenciés.
