An improved system for screening a multiple of candidate therapeutic or chemotherapeutic agents for efficay as to a specific patient, in which a tissue sample from the patient is harvested, cultured and separately exposed to a plurality of treatments and/or therapeutic agents for the purpose of objectively identifying the best treatment or agent for the particular patient. Specific method innovations such as tissue sample preparation techniques render this method practically as well as theoretically useful. One particularly important tissue sample preparation technique is the initial preparation of cohesive multicellular particulates of the tissue sample, rather than enzymatically dissociated cell suspensions or preparations, for initial tissue culture monolayer preparation. By subjecting uniform samples of cells to a wide variety of active agents (and concentrations thereof), the most promising agent and concentration for treatment of a particular patient can be determined.
L'invention concerne un système amélioré permettant d'examiner plusieurs agents thérapeutiques ou chimiothérapeutiques candidats en ce qui concerne leur efficacité vis-à-vis d'un patient spécifique. Selon l'invention, un échantillon tissulaire est prélevé sur le patient, mis en culture et exposé séparément à une pluralité de traitements et/ou d'agents thérapeutiques, cela pour permettre d'identifier objectivement le meilleur traitement ou agent pour le patient particulier. Des innovations de procédé spécifiques, telles que des techniques de préparation de l'échantillon tissulaire, font que ce procédé est utile tant sur le plan pratique que sur le plan théorique. Une technique de préparation de l'échantillon tissulaire particulièrement importante est la préparation initiale de substances particulaires, multicellulaires, cohérentes, constituées de l'échantillon tissulaire, plutôt que l'utilisation de préparations ou de suspensions cellulaires dissociées enzymatiquement, pour la préparation d'une couche simple de culture tissulaire initiale. On peut, en soumettant des échantillons uniformes de cellules à une large variété d'agents actifs (et de concentrations de ces agents actifs), déterminer l'agent ou la concentration susceptible de donner le meilleur résultat pour le traitement d'un patient particulier.
