The present invention provides improved cells and methods for identifying compounds that alter protein expression, such as xenobiotics, endobiotics, chemical or drugs. One aspect of the present invention is a cell that includes a first nucleic acid molecule that includes: a promoter or enhancer operable for a nucleic acid molecule encoding a protein involved in drug metabolism (such as an enzyme or transporter) and a reporter gene; and a second nucleic acid encoding an intracellular receptor or transcription factor; so that when the intracellular receptor or transcription factor is bound with a compound, the intracellular receptor or transcription factor can operably bind with the promoter or enhancer resulting in the expression of said reporter gene. Another aspect of the present invention is a method for evaluating compounds for the property of inducing the expression of a gene encoding a protein involved in drug metabolism, including; providing a test compound; contacting the test compound with a cell of the present invention; and detecting the expression of said reporter gene. This method can be a high throughput method.
L'invention concerne des cellules et procédés perfectionnés servant à identifier des composés qui modifient l'expression de protéines, tels que des xénobiotiques, endobiotiques, substances chimiques ou médicaments. L'invention comporte également d'autres aspects avantageux, dont l'un notamment porte sur une première molécule d'acides nucléiques comprenant un promoteur ou activateur pouvant être mis en oeuvre pour une molécule d'acides nucléiques codant pour une protéine impliquée dans le métabolisme d'un médicament (telle qu'une enzyme ou un transporteur), ainsi qu'un gène marqueur; ainsi que sur une seconde molécule d'acides nucléiques codant pour un récepteur intracellulaire ou facteur de transcription; de sorte que, lorsque le récepteur intracellulaire ou facteur de transcription est lié à un composé, le récepteur intracellulaire, transporteur ou facteur de transcription peut se lier de manière opérationnelle au promoteur ou activateur, induisant ainsi l'expression du gène marqueur. Un autre aspect de l'invention concerne un procédé d'évaluation de composés, destiné à mettre en évidence la propriété d'induction de l'expression d'un gène codant pour une protéine impliquée dans le métabolisme d'un médicament, ce procédé consistant à préparer un composé d'essai, à mettre en contact ce composé d'essai avec une cellule de l'invention, et à détecter l'expression du gène marqueur. Le procédé de l'invention peut constituer un procédé à rendement élevé.
