An improved process, and intermediates, for the preparation of 4'- trifluoromethyl-biphenyl-2-carboxylic acid (1,2,3,4-tetrahydro-isoquinolin-6- yl)-amide (I). Compound (I) is useful in the preparation of 4'-trifluoromethyl- biphenyl-2-carboxylic acid [2-(1H-[1,2,4-triazol-3-ylmethyl)]-(1,2,3,4- tetrahydro-isoquinolin-6-yl)-amide (II) which is useful as an inhibitor of microsomal triglyceride transfer protein and/or apolipoprotein B (Apo B) secretion, and which are accordingly useful for the preparation and treatment of atherosclerosis and its clinical sequelae, for lowering serum lipids, and related diseases. A compound of the formula A-CHR1R2 wherein A is a group of formula (XII) and R1 selected from H and OR3 wherein R3 is hydrogen or (C1- C6)alkyl; R2 is selected from OR3, -CH2-NH2.(R5H)m, wherein m and R5H are defined below, and -CH2-NO2; or R1 and R2 together form an =O or =CH-NO2 group; with the proviso that when R1 and R2 are each an OR3 group then both R3s are (C1-C6)alkyl groups and the same.
Cette invention se rapporte à un procédé perfectionné, et aux intermédiaires associés, de préparation de 4'-trifluorométhyl-biphényl-2-acide carboxylique (1,2,3,4-tétrahydro-isoquinoline-6-yl)-amide (I). Le composé (I) est utile à la préparation de 4'-trifluorométhyl-biphényl-2-acide carboxylique [2-(1H-[1,2,4-triazole-3-ylméthyl)]-(1,2,3,4-tétrahydro-isoquinoline-6-yl)-amide (II), composé qui s'avère utile en tant qu'inhibiteur de sécrétion des protéines de transfert de triglycéride microsomal et/ou d'apolipoprotéines B (Apo B), et qui est par conséquent utilisé pour prévenir et traiter l'athérosclérose et ses séquelles cliniques, pour réduire le taux de lipides sériques et pour traiter les maladies associées. L'invention se rapporte en outre à un composé de formule A-CHR?1¿R?2¿ dans laquelle A est un groupe représenté par la formule (XII) et R?1¿ est sélectionné parmi H et OR?3¿, où R?3¿ est hydrogène ou alkyle (C¿1?-C¿6?); R?2¿ est sélectionné parmi OR?3¿, -CH¿2?-NH¿2? (R?5¿H)¿m?, m et R?5¿H étant définis ci-dessus, et -CH¿2?-NO¿2?; ou R?1¿ et R?2¿ forment conjointement un groupe =O ou =CH-NO¿2?; à condition que lorsque R?1¿ et R?2¿ sont chacun OR?3¿, alors ces groupes R?3¿ sont tous deux des groupes alkyle (C¿1?-C¿6?) et analogues.
