Abstract The present invention relates to recombinant methods for producing a secreted polypeptide having L-asparaginase activity, comprising (a) cultivating under conditions conducive for production of the polypeptide a host cell comprising a nucleic acid construct comprising a first nucleic acid sequence encoding a secretory signal peptide operably linked to second nucleic acid sequence encoding the polypeptide having L-asparaginase activity, wherein the signal peptide directs the polypeptide into the cell's secretory pathway; and (b) recovering the secreted polypeptide. The present invention also relates to isolated polypeptides having L-asparaginase activity and nucleic acids thereof.
L'invention concerne des procédés de recombinaison, destinés à produire un polypeptide sécrété à activité L-asparaginase, consistant (a) à cultiver, dans des conditions favorables à la production du polypeptide, une cellule hôte comprenant une construction d'acides nucléiques comportant une première séquence d'acides nucléiques, codant pour un peptide du signal sécrétoire, liée de façon opératoire à une seconde séquence d'acides nucléiques, codant pour le polypeptide à activité L-asparaginase, le peptide du signal dirigeant le polypeptide dans la voie sécrétoire de la cellule, et (b) à récupérer le polypeptide sécrété. L'invention concerne aussi des polypeptides isolés à activité L-asparaginase et des acides nucléiques s'y rapportant.
