A 17kb nucleic acid fragment, which confers production of the lantibiotic cinnamycin on non-producer Streptomycete strains, has been isolated from Streptomyces cinnamoneus cinnamoneus DSM 40005 and characterised. Also provided are variants of the 17kb fragment including variants in which non- essential genes are deleted and variants in which the propeptide sequence of the cinnamycin structural gene is altered, for example replaced with the propeptide sequence of similar B-type lantibiotics. Also provided are vectors, host cells and methods of lantibiotic production using the nucleic acid of the invention, and libraries of variants.
L'invention concerne un fragment d'acide nucléique de 17 kb, permettant de produire le lantibiotique cinnamycine sur des souches de Streptomycète non productrices, ayant été isolées à partir de Streptomyces cinnamoneus cinnamoneus DSM 40005, et caractérisées. L'invention concerne également des variants du fragment de 17 kb comprenant des variants dans lesquels des gènes non essentiels sont supprimés, et des variants dans lesquels la séquence propeptidique du gène structural de cinnamycine est modifiée, par exemple remplacée par la séquence propeptidique de lantibiotiques de type B similaires. L'invention concerne encore des vecteurs, des cellules hôtes, et des procédés de production de lantibiotique faisant appel à l'acide nucléique de l'invention, et à des bibliothèques de variants.
