Various system and method embodiments of the present invention are directed to separating target molecules from complex solutions by affinity column chromatography using organic-solvent-containing eluants. In one embodiment of the present invention, an eluant containing an organic-solvent is used, at a first pH, to remove non-target solutes and suspended entities from an affinity chromatography column. The pH of the eluant is then changed to a second pH, and the organic- solvent-containing eluant is used to elute target molecules from the affinity column chromatography.

La présente invention concerne divers modes de réalisation de systèmes et de procédés destinés à séparer des molécules cibles de solutions complexes par chromatographie d~affinité sur colonne à l~aide d~éluants contenant des solvants organiques. Selon un mode de réalisation de la présente invention, un éluant contenant un solvant organique est utilisé, à un premier pH, pour retirer des solutés non-cibles et des entités en suspension d~une colonne de chromatographie d~affinité. Le pH de l~éluant est ensuite changé en un second pH et l~éluant contenant un solvant organique est utilisé pour éluer les molécules cibles de la chromatographie d~affinité sur colonne.
