The present invention provides methods for reducing b-amyloid deposition, b- amyloid neurotoxicity and microgliosis in animals or humans afflicted with a cerebral amyloidogenic disease, such as Alzheimer's disease (AD), by administering therapeutically effective amounts of the dihydropyridine calcium channel antagonist, nilvadipine. The present invention also provides methods for diagnosing cerebral amyloidogenic diseases in animals or humans. Further provided are methods for reducing the risk of b-amyloid deposition, b-amyloid neurotoxicity and microgliosis in animals or humans suffering from traumatic brain injury by administering nilvadipine immediately after the traumatic brain injury and continuing treatment for a prescribed period of time thereafter. Finally, methods are provided for treating transplantable neuronal stem cells by administering nilvadipine to the neuronal stem cells prior to transplantation in the central nervous system of an animal or human afflicted with a cerebral amyloidogenic disease, such as AD.

La présente invention concerne des méthodes de réduction d'un dépôt b-amyloïde, d'une neurotoxicité b-amyloïde ou d'une microgliose chez des animaux ou des humains atteints d'une maladie amyloïdogénique cérébrale, telle que la maladie d'Alzheimer (MA), qui consistent à administrer des quantités thérapeutiquement efficaces d'un antagoniste des canaux calciques de type dihydropyridine, le nilvadipine. L'invention concerne également des méthodes destinées à diagnostiquer des maladies amyloïdogéniques cérébrales chez des animaux ou des humains, et des méthodes destinées à réduire le risque de dépôt b-amyloïde, de neurotoxicité b-amyloïde et de microgliose chez des animaux ou des humains atteints d'une lésion cérébrale traumatique, par l'administration de nilvadipine immédiatement après le traumatisme cérébral et la poursuite du traitement pendant une durée consécutive prescrite. L'invention concerne en outre des méthodes de traitement de cellules souches neuronales transplantables, par administration de nilvadipine aux cellules souches neuronales préalablement à la greffe dans le système nerveux central d'un animal ou d'un humain atteint d'une maladie amyloïdogénique cérébrale comme la MA.
