The present invention relates to a process for synthesizing or amplifying efficiently a nucleic acid comprising a target nucleic acid sequence. In the process according to the present invention, a primer comprising in its 3'-end portion a sequence (Ac') which hybridizes a sequence (A) in the 3'-end portion of the target nucleic acid sequence, and in the 5'-side of said sequence (Ac') a sequence (B') which hybridizes the complementary sequence (13c) of a sequence (B) positioned in the 5'-side of said sequence (A) on the target nucleic acid sequence, wherein {X - (Y - Y')}/X is in the range of -1.00 to 1.00, in which X denotes the number of bases in said sequence (Ac'), Y denotes the number of bases in the region flanked by said sequences (A) and (B) in the target nucleic acid sequence, and Y' denotes the number of bases in an intervening sequence between said sequences (Ac') and (B') (Y' may be zero).

L'invention concerne une méthode permettant de synthétiser ou d'amplifier efficacement un acide nucléique contenant une séquence d'acide nucléique cible. Dans cette méthode, on utilise une amorce qui contient dans sa partie 3'-terminale une séquence (Ac') hybridable avec la séquence 3'-terminale (A) d'une séquence d'acide nucléique cible et qui contient également dans sa partie 5'-latérale une séquence (B') hybridable avec une séquence (Bc) complémentaire d'une séquence (B) située dans le côté 5' de la séquence (A) dans la séquence d'acide nucléique cible. Dans ladite amorce, {X-(Y-Y')}/X est compris entre 1,00 et 1,00, X représentant le nombre de bases comprises dans la séquence (Ac'), Y représentant le nombre de bases comprises dans la région située entre la séquence (A) et la séquence (B) dans la séquence d'acide nucléique et Y' représentant le nombre de bases comprises dans la séquence d'insertion comprise entre la séquence (Ac') et la séquence (B'), à la condition que Y' puisse valoir 0.
