Platelet-specific, chimeric immunoglobulin and immunoglobulin fragments are described. The chimeric molecules are made up of a nonhuman antigen binding region and a human constant region. Preferred immunoglobulins are specific for glycoprotein IIb/IIIa receptor in its complexed form; they block ligand binding to the receptor and prevent platelet aggregation. The immunoglobulins can be used to reduce or prevent occlusion, reocclusion (e.g, abrupt closure), stenosis and/or restenosis. In addition, the immunoglobulins are useful in anti-thrombotic therapy when administered alone or in conjunction with thrombolytic agents, as well as in thrombus imaging.

L'invention concerne l'immunoglobuline chimérique spécifique des plaquettes et des fragments d'immunoglobuline. Les molécules chimériques se composent d'un site non humain de combinaison avec l'antigène et d'un site humain constant. Les types d'immunoglobuline préférés sont spécifiquement dirigés contre le récepteur de glycoprotéine IIb/IIIa dans sa forme complexe. Ils bloquent la combinaison du ligand avec le récepteur et préviennent l'agrégation plaquettaire. Les immunoglobulines s'utilisent pour réduire ou prévenir l'occlusion, la réocclusion (c.à.d. la fermeture subite), la sténose et/ou la sténose récidivante. De plus, les immunoglobulines s'utilisent dans le traitement antithrombotique lorsqu'elles sont administrées seules ou conjointement avec des agents antithrombolytiques, de même que dans les procédés d'imagerie permettant la visualisation des thrombus.
