A methodology that allows for highly efficient transfer and stable integration of DNA into both established eukaryotic cell lines and primary cells, including non-dividing cells such as human peripheral blood monocytes and macrophages entails the use of a synthetic polypeptide comprised of a peptide domain which corresponds to a nuclear localization signal sequence and a DNA binding domain which is rich in basic amino acids, separated by a hinge region of neutral amino acid which prevents stearic interference between the two domains.

L'invention concerne une méthode permettant un transfert très efficace et une intégration stable d'ADN dans le génome de cellules eucaryotiques de lignées établies ou de cellules primaires telles que les cellules incapables de se diviser comme les monocytes et les macrophages du sang périphérique. Cette méthode fait appel à un polypeptide de synthèse comprenant un domaine peptidique correspondant à une séquence de signal de localisation nucléaire et un domaine de fixation à l'ADN, qui est riche en acides aminés basiques, ces deux domaines étant séparés par une région charnière d'acides aminés neutres qui empêche les interférences stériques entre les deux domaines.
