An integrative DNA vector and one or more viral proteins having affinity for DNA are packaged in cochleate precipitates. The integrative DNA vector contains one or more therapeutic nucleotide sequences that are preferably positioned between DNA substrates for the proteins. Upon contact with a lipid bilayer of a target cell, the cochleate vector structure delivers one or more of the therapeutic nucleotide sequences and one or more proteins to the interior of the target cell. Upon entry into the cell, the proteins facilitate the integration of the therapeutic nucleotide sequence into the genome of the host cell.
L'invention concerne un vecteur ADN d'intégration et au moins une protéine virale présentant une affinité pour l'ADN encapsulés dans des précipités cochléaires. Ledit vecteur contient au moins une séquence nucléotidique thérapeutique placée, de préférence, entre des substrats d'ADN pour les protéines. Au contact avec une bicouche lipidique d'une cellule cible, la structure du vecteur cochléaire délivre au moins une séquence nucléotidique thérapeutique et au moins une protéine à l'intérieur de la cellule cible. Dès l'entrée dans la cellule, les protéines facilitent l'intégration de la séquence nucléotidique thérapeutique dans le génome de la cellule hôte.
