Human and rat cAMP phosphodiesterase isoforms (denoted PDE4Ds), as well as the DNA (RNA) encoding such polypeptides, are disclosed. Also disclosed are methods for utilizing such polypeptides in diagnostic assays for identifying mutations in nucleic acid sequences encoding the polypeptides of the present invention, for detecting altered levels of the polypeptide of the present invention as a means of detecting diseases and methods of screening potential modulators, especially inhibitors, of the PDE4Ds (denoted PDE4D6) disclosed herein as a means of increasing cyclic AMP in neurons and thus treating neurological problems, such as long term memory loss, if not preventing such maladies entirely. Transgenic animals expressing polypeptides disclosed herein are also described.
La présente invention concerne des isoformes de la phosphodiestérase du cAMP de l'homme et du rat (désignées par PDE4Ds), ainsi que l'ADN (ARN) codant pour de tels polypeptides. Cette invention concerne également des procédés d'utilisation de tels polypeptides dans des méthodes diagnostiques servant à l'identification de mutations dans des séquences d'acide nucléique codant pour les polypeptides de l'invention, des procédés de détection d'états altérés du polypeptide de l'invention servant à la détection de maladies, et des procédés de criblage de modulateurs potentiels, particulièrement d'inhibiteurs, des PDE4Ds (désignées par PDE4D6) permettant, d'après l'invention, d'augmenter l'AMP cyclique dans les neurones et donc de traiter les problèmes neurologiques tels que les pertes de mémoire à long terme, si ce n'est de prévenir complètement l'apparition de telles maladies. Cette invention concerne également des animaux transgéniques exprimant les polypeptides en question.
