Cells suffering from the deletion or inhibition of the function of expressing a normal gene (wild type gene) in alleles corresponding to a trapped gene which are applicable also to in vitro cell differentiation systems other than ES cells; a method of preparing the same; a gene trap method with the use of the above cells; and an antisense promoter transfer vector to be used in the preparation of the above cells. By transferring the gene trap vector into cells, a trap clone can be efficiently obtained. By inserting a powerful promoter into a definite position in a mutated locus, wherein the insertion of the gene trap vector of the trapped clone has occurred, in the direction opposite to the endogenous gene, moreover, the antisense RNA against the trapped gene is compulsively transcribed and thus the transcription product from the wild type gene is disrupted.

L'invention concerne des cellules souffrant de la délétion ou de l'inhibition de la fonction d'expression d'un gène normal (gène de type sauvage) dans des allèles correspondant à un gène piégé, qui sont également applicables à des systèmes de différentiation de cellules <i>in vitro</i> autres que des cellules ES. L'invention concerne également un procédé permettant de les préparer; un procédé de piégeage génique utilisant lesdites cellules; et un vecteur de transfert de promoteur à utiliser dans la préparation desdites cellules. En transférant le vecteur de piégeage génique dans les cellules, on obtient de manière efficace un clone-piège. En introduisant, dans un locus muté, un promoteur puissant suivant une position déterminée, où l'introduction du vecteur de piégeage génique du clone piégé est survenue, dans la direction opposée au gène endogène, l'ARN antisens contre le gène piégé étant en outre transcrit par contrainte, on interrompt, par conséquent, le produit de transcription à partir du gène de type sauvage.
