Methods for the production of monoclonal antibodies specific to conformational epitope(s) of a prefibrilar aggregate(s) which contribute to amyloid fibril formation in human or animal subjects who suffer from amyloid diseases (e.g. Alzheimer's Disease) and the hybridomas and monoclonal antibodies produced therefrom. Also, the use of such monoclonal antibodies in the immunization of human or animal subjects against Alzheimer's Disease or other amyloid diseases and/or for the diagnosis or detection of Alzheimer's Disease or other amyloid diseases. The monoclonal antibodies may be administered concomitantly or in combination with anti-inflammatory agents, such as gold or gold containing compounds, to decrease neural inflammation associated with amyloid diseases (e.g. Alzheimer's Disease).
L'invention porte sur des procédés de production d'anticorps monoclonaux spécifiques à un ou des épitopes conformationnels d'un ou d'agrégats préfibrilaires qui contribuent à la formation de fibrilles amyloïdes chez des êtres humains ou des animaux souffrant de maladies amyloïdes (comme la maladie d'Alzheimer) et sur les hybridomes et les anticorps monoclonaux obtenus. L'invention porte également sur l'utilisation de ces anticorps monoclonaux dans l'immunisation des êtres humains ou des animaux contre la maladie d'Alzheimer ou autres maladies amyloïdes et/ou dans le diagnostic ou la détection de la maladie d'Alzheimer ou autres maladies amyloïdes. Les anticorps monoclonaux peuvent être administrés simultanément ou en combinaison avec des agents anti-inflammatoires, tels que de l'or ou des composés contenant de l'or, afin de diminuer les inflammations neuronales associées à des maladies amyloïdes (telles que la maladie d'Alzheimer).
