The present provides a mammal in which the expression of one or more interleukin genes has been suppressed. More specifically, the invention concerns the inactivating deletion of the interleukin-1 receptor antagonist gene to produce a knock-out non-human mammal with decreased or completely suppressed expression of the endogenous gene. The invention provides methods for preparing such knock-out mammals and methods of using the knock-out mammals to evaluate the effectiveness of therapeutic agents and regimens to treat diseases or disorders associated with perturbations in the interleukin pathways.

La présente invention concerne un mammifère dans lequel l'expression d'un ou plusieurs gènes d'interleukine a été supprimée. En particulier, l'invention concerne la délétion inactivante du gène antagoniste du récepteur de l'interleukine-1 de manière à produire un mammifère non humain dont on a réduit ou complètement supprimé l'expression du gène endogène. L'invention concerne des procédés de mise au point de tels mammifères "inactivés", ainsi que des procédés d'utilisation de ces mammifères "inactivés" pour évaluer l'efficacité d'agents thérapeutiques et de schémas de posologie dans le traitement des maladies ou des troubles associés à des perturbations des voies d'interleukines.
