Three genes encoding membrane-secreted proteins have been successfully isolated from osteoblast-like cell lines by using a method for specifically cloning membrane-secreted proteins. As the result of subsequent analysis on one of these genes, it has been found out that the protein encoded by this gene is a novel receptor protein which has an extracellular region alone, binds to cell membranes via a GPI anchor and carries therein a cysteine-rich repetitive region conserved in the TNF receptor super family. It has been also found out that when this protein is largely expressed in an osteoblast-like cell line, the proliferation of the cells are suppressed, the cells are morphologically changed, and the alkaline phosphatase activity which is one of the indications of the differentiation of osteoblasts is elevated.
On a réussi à isoler de lignées cellulaires de type ostéoblastes trois gènes codant des protéines sécrétées par une membrane à l'aide d'un procédé de clonage spécifique des protéines sécrétées par une membrane. Après avoir analysé un de ces gènes, on a découvert que la protéine codée par ce gène est une nouvelle protéine réceptrice qui comporte une région extracellulaire unique, qui se fixe aux membranes cellulaires par l'intermédiaire d'une molécule GPI ancrée et supporte dans celle-ci une région répétitive riche en cystéine conservée dans la super famille du récepteur du facteur de nécrose tumorale (TNF). On a également découvert que, lorsque cette protéine est en grande partie exprimée dans une lignée cellulaire de type ostéoblastes, la prolifération des cellules est réprimée, les cellules sont modifiées morphologiquement et l'activité de la phosphatase alcaline, qui est une des indications de la différentiation des ostéoblastes, est élevée.
