The invention concerns a family of peptides exhibiting high affinity and specificity for the gp120 viral protein, and methods for producing said peptides, and the use of said peptides. The inventive peptides are characterised in that they comprise the following sequence (A): TPA-P1-Cys-P2- Cys-P3-Cys-Ala or Gln-Gly or (D) Asp or Ser or His or Asn-XaaJ-Cys Thr or Ala- Cys-Xaak-NH2, wherein TPA represents thiopropionic acid; XaaJ represents .beta.-naphthylalanine, phenylalanine or bi-phenylalanine; Xaak represents Gly, Val or Ileu; P1 represents 3 to 6 amino acids, P2 represents 2 to 4 amino acids and P3 represents 6 to 10 amino acids, the amino acids in P1, P2 and P3 being natural or synthetic, identical or different and P1, P2 and P3 having or not having a common sequence, said peptide having a .beta. hairpin conformation whereof the .beta. bend is formed by Ala or Glu-Gly or CDAsp or Ser or His or Asn-Xaak amino acid residues of its sequence (A). Said peptide can be used for making a medicine, a vaccine, a chromatography column and for screening.
La présente invention se rapporte à une famille de peptides présentant une affinité et une spécificité élevées pour la protéine virale gp120, à des procédés de fabrication de ces peptides, et à l'utilisation de ces peptides. Le peptide de la présente invention se caractérise en ce qu'il comprend la séquence (A) suivante : TPA - P?1¿ - Cys - P?2¿ - Cys - P?3¿ - Cys - Ala ou Gln -Gly ou (D) Asp ou Ser - Ser ou His ou Asn - Xaa?J¿ - Cys Thr ou Ala - Cys- Xaa?k¿ - NH¿2? dans laquelle TPA représente l'acide thiopropionique, Xaa?J¿ représente la .beta.-napthylalanine, la phénylalanine ou la bi-phénylalanine, Xaa?k¿ représente Gly, Val ou Ileu, P?1¿ représente 3 à 6 acides aminés, P?2¿ représente 2 à 4 acides aminés et P?3¿ représente 6 à 10 acides aminés, les acides aminés dans P?1¿, P?2¿ et P?3¿ étant naturels ou non naturels, identiques ou différents et P?1¿, P?2¿ et P?3¿ ayant ou non une séquence commune, ledit peptide présentant une conformation en épingle à cheveux .beta. dont le coude .beta. est formé par les résidus acides aminés Ala ou Glu - Gly ou CDAsp ou Ser ou His ou Asn- Xaa?k¿.de sa séquence (A). Il peut être utilisé pour fabriquer un médicament, un vaccin, une colonne de chromatographie et pour faire un criblage.
