The invention relates to a method For purification of a mixture of hydroxamate derivatized protein and native protein which is characterized by treating the mixture with immobilized metal and thereby adsorbing the hydroxamate derivatized protein/protein on the immobilized-metal and recovering the native protein. The protein could be IGF-I. It also relates to a process for the production of a native protein which is characterized by expression of the protein as a fusion protein, cleavage of the fusion protein by hydroxylamine separation of native protein from hydroxamate derivatized protein by adsorbing the hydroxamate derivatized protein on immobilized metal and directly recovering the native protein. The use of immobilized metal affinity chromatography for separation of native protein from hydroxamate derivatized protein is also claimed.
L'invention concerne un procédé pour la purification d'un mélange d'une protéine/de protéines dérivée(s) de l'hydroxamate et d'une protéine à l'état naturel. Ce procédé se caractérise par le traitement du mélange avec du métal immobilisé, puis par l'adsorption de la protéine/de protéines dérivée(s) de l'hydroxamate sur le métal immobilisé et la récupération de la protéine à l'état naturel. La protéine peut être l'IGF-I. L'invention a aussi pour objet la production d'une protéine à l'état naturel qui est caractérisée par les étapes suivantes: expression de la protéine sous forme d'une protéine de fusion, scission de la protéine de fusion par l'hydroxylamine, séparation de la protéine à l'état naturel de la protéine dérivée de l'hydroxamate réalisée par adsorption de la protéine dérivée de l'hydroxamate sur le métal immobilisé et récupération directe de la protéine à l'état naturel. L'invention utilise également la chromatographie d'affinité pour le métal immobilisé pour séparer la protéine à l'état naturel de la protéine dérivée de l'hydroxamate.
