The invention features a method for the selection and expansion of bone marrow stromal cells. The method includes the steps of obtaining bone marrow stromal cells, introducing the stromal cells into a vessel precoated on an inner surface with a gelatin, and containing a culture medium including an acidic fibroblast growth factor ("aFGF") polypeptide, and expanding the stromal cells in the culture medium under conditions and for a time sufficient to obtain an increased number of bone marrow stromal cells. The culture medium additionally can include heparin, and the vessel additionally can be precoated with fetal bovine serum.
Cette invention concerne un procédé de sélection et d'expansion de cellules de stroma de moelle osseuse, lequel procédé comprend les étapes suivantes: obtenir des cellules de stroma de moelle osseuse; introduire ces cellules de stroma dans un récipient dont on a préalablement revêtu une surface interne de gélatine, et qui contient un milieu de culture comprenant un polypeptide de facteur de croissance de fibroblaste acide ("aFGF"); et enfin, effectuer l'expansion des cellules de stroma dans le milieu de culture dans des conditions et pendant une durée qui permettent d'obtenir un plus grand nombre de cellules de stroma.
