The present invention relates to methods for obtaining a polypeptide construct directed against one or more antigens and/or epitopes and having one or more desired characteristics, wherein the polypeptide construct comprises at least two single domain antibodies. The methods of the present invention involve producing a diversity of polypeptide constructs that are structural variants and screening the produced diversity of polypeptide constructs for a polypeptide construct having said one or more desired characteristics. The present invention further relates to polypeptide constructs directed against one or more antigens and/or epitopes having one or more desired characteristics, wherein the polypeptide construct comprises at least two single domain antibodies. The methods and polypeptide constructs according to the present invention are useful for the identification of optimal therapeutic compounds.
La présente invention porte sur des procédés pour obtenir un produit de construction de polypeptide dirigé contre un ou plusieurs antigènes et/ou épitopes et ayant une ou plusieurs caractéristiques désirées, le produit de construction de polypeptide comprenant au moins deux anticorps à domaine unique. Les procédés de la présente invention mettent en jeu la production d'une diversité de produits de construction de polypeptide qui sont des variants structuraux et le criblage de la diversité produite de produits de construction de polypeptide pour un produit de construction de polypeptide ayant ladite ou lesdites caractéristiques désirées. La présente invention porte en outre sur des produits de construction de polypeptide dirigés contre un ou plusieurs antigènes et/ou épitopes ayant une ou plusieurs caractéristiques désirées, le produit de construction de polypeptide comprenant au moins deux anticorps à domaine unique. Les procédés et produits de construction de polypeptide selon la présente invention sont utiles pour l'identification de composés thérapeutiques optimaux.
