The present invention relates to the use of a peptide composition as an immunogen, with each peptide contained therein comprising a target antigenic site derived from the VP1 capsid protein of Foot-and-Mouth Disease Virus (FMDV). The antigenic site is covalently linked to a helper T cell epitope and, preferably, to other immunostimulatory sequences, preferably by conventional peptide bond(s) through direct synthesis, for the prevention of FMDV infection and eradication of Food-and-Mouth Disease (FMD). More particularly, the present invention relates to the use of such peptide composition as an immunogen to elicit the production in animals including swine, cattle, sheep, goets and susceptible wild species, of high titer polyclonal antibodies that can effectively neutralize, in vitro, multiple strains or serotypes of FMDV, and to the use of such composition as a vaccine to prevent, and/or reduce the incidence of, FMDV infection regardless of serotype, and thus affect the eradication of FMD. The present invention also relates to the peptides used in the compositions, and to immunoassays and/or diagnostic kits containing one or more of these peptides, and methods of diagnosing FMDV in mammals using such materials.

L'invention concerne l'utilisation d'une composition peptidique comme immunogène. Chaque peptide contenu dans cette composition comprend un site antigénique cible dérivé de la protéine à capside VP1 du virus de la fièvre aphteuse. Le site antigénique est lié par covalence à un épitope de lymphocyte T auxiliaire et de préférence à d'autres séquences immunostimulatrices, de préférence par liaison peptidique classique au moyen d'une synthèse directe, afin de prévenir une infection au virus de la fièvre aphteuse et d'éradiquer celui-ci. D'une manière spécifique, l'invention concerne l'utilisation de cette composition peptidique en tant qu'immunogènes pour déclencher, chez les animaux tels que le porc, le boeuf, le mouton, la chèvre et éventuellement, des espèces sauvages, la production d'anticorps polyclonaux à titre élevé pouvant efficacement neutraliser in vitro de multiples souches ou sérotypes de virus de la fièvre aphteuse. L'invention concerne également, d'une manière spécifique, l'utilisation de cette composition comme vaccin pour prévenir et/ou réduire la survenance d'une infection au virus de la fièvre aphteuse sans égard au sérotype et ainsi permettre l'éradication de la fièvre aphteuse. L'invention concerne également des peptides utilisés dans lesdites compositions, un dosage immunologique et/ou des trousses de diagnostic contenant un ou plusieurs de ces peptides ainsi que des méthodes permettant de diagnostiquer au moyen de ces matériaux le virus de la fièvre aphteuse chez des mammifères.
