Methods of preparing recombinant Bacillus anthracis protective antigen or a variant or fragment thereof for use in vaccines is disclosed. The protein is expressed in a recombinant microorganism which comprises a sequence which encodes PA or said variant or fragment thereof wherein either (i) a gene of the microorganism which encodes a catabolic repressor protein and/or AbrB is inactivated, and/or (ii) wherein a region of the PA sequence which can act as a catabolic repressor binding site and/or an AbrB binding site is inactivated. Useful quantities of protein are obtainable from these organisms.

L'invention porte sur des procédés de préparation d'un antigène protecteur contre la bactéridie charbonneuse, d'une variante ou d'un fragment de celui-ci, à utiliser dans des vaccins. La protéine est exprimée dans un micro-organisme recombiné comprenant une séquence codant pour l'antigène protecteur, ladite variante, ou encore ledit fragment de celui-ci, dans lequel soit i) un gène dudit micro-organisme codant pour un répresseur catabolique et/ou AbrB est inactivé, soit ii) une zone de la séquence d'antigène protecteur peut agir en tant que site de liaison de répresseur catabolique et/ou un site de liaison AbrB est inactivé, soit les deux phénomènes i) et ii) se produisent. Des quantités utiles de protéines peuvent être produites à partir de ces organismes.
