A human TNF-gamma polypeptide and DNA (RNA) encoding such polypeptide and a procedure for producing such polypeptide by recombinant techniques is disclosed. Also disclosed are methods for utilizing such polypeptide to inhibit cellular growth, for example in a tumor or cancer, for facilitating wound-healing, to provide resistance against infection, induce inflammatory activities, and stimulating the growth of certain cell types to treat diseases, for example restenosis. Also disclosed are diagnostic methods for detecting a mutation in the TNF-gamma nucleic acid sequence or an overexpression of the TNF-gamma polypeptide. Antagonists against such polypeptides and their use as a therapeutic to treat cachexia, septic shock, cerebral malaria, inflammation, arthritis and graft-rejection are also disclosed.

Cette invention concerne un polypeptide gamma du type facteur de nécrose tumorale (FNT) d'origine humaine et l'ADN (ARN) de codage de ce polypeptide ainsi qu'une procédure de production dudit polypeptide à l'aide de techniques de recombinaison. Des procédés d'utilisation du polypeptide pour inhiber la croissance cellulaire, dans une tumeur ou un cancer par exemple, en vue de faciliter la cicatrisation des plaies, de conférer une résistance contre les infections, d'induire des activités inflammatoires et de stimuler la croissance de certains types de cellules pour traiter des maladies, la resténose par exemple, sont également décrits, ainsi que des procédés de diagnostic qui permettent de détecter une mutation dans la séquence nucléotidique du polypeptide gamma-FNT ou une surexpression du polypeptide gamma-FNT, et, pour terminer, des antagonistes dirigés contre les polypeptides de cette invention et leur utilisation comme agents thérapeutiques pour traiter la cachexie, le choc septique, le paludisme cérébral, les inflammations, l'arthrite et le rejet des greffes.
