An isolated clone consisting of sequences transcribed from the TNF-gamma gene. Also provided are human polypeptides translated from said TNF-gamma sequences and a procedure for producing such polypeptide by recombinant techniques. Also provided are a procedure for producing soluble biologically active TNF-gamma, which may be used to treat deficiencies of TNF-gamma and diseases conditions ameliorated by TNF-gamma. Antibodies, antagonists and inhibitors of such polypeptide which may be used to prevent the action of such polypeptide and therefore may be used therapeutically to treat TNF-gamma associated diseases, tumors or metastases are disclosed. Also disclosed is the use of said antibodies, agonists and inhibitors as well as the nucleic acid sequences to screen for, diagnose, prognosticate, stage and monitor conditions and diseases attributable to TNF-gamma, especially inflammation. The use of said partial sequence to provide antibodies, agonists and inhibitors as well as partial nucleic acid sequences to screen for, diagnose, stage and monitor diseases associated with TNF-gamma, including but not limited to inflammation. Illustrative sequences and clone designations for TNF-gamma are provided.
L'invention concerne un clone isolé formé de séquences transcrites à partir du gène de TNF-gamma. L'invention concerne aussi des polypeptides humains traduits depuis lesdites séquences de TNF-gamma et une procédure relative à l'élaboration de ce type de polypeptide par des techniques de recombinaison. L'invention concerne également une procédure relative à l'élaboration de TNF-gamma actif biologiquement soluble, susceptible d'être utilisé pour traiter les déficiences de TNF-gamma et les affections dont l'état est amélioré par le TNF-gamma. L'invention concerne en outre des anticorps, des antagonistes et des inhibiteurs des polypeptides considérés, utilisables pour prévenir l'action de ce type de polypeptide, et donc thérapeutiquement utilisables pour traiter les maladies, les tumeurs ou les métastases associées au TNF-gamma. On décrit aussi l'utilisation des anticorps, agonistes et inhibiteurs en question ainsi que les séquences nucléotidiques permettant le dépistage, le diagnostic, le pronostic, la stadification et le contrôle des maladies imputables au TNF-gamma, en particulier l'inflammation. On décrit en outre l'utilisation de la séquence partielle considérée pour fournir des anticorps, des agonistes et des inhibiteurs, et les séquences nucléotidiques partielles permettant le dépistage, le diagnostic, la stadification et le contrôle des maladies asociées au TNF-gamma, y compris mais pas seulement l'inflammation.
