A composition for administration of a therapeutic compound to a multi-drug resistant cell in a person suffering from a drug-resistant cancer is described. The composition is composed of a carrier molecule and a folate targeting ligand, which is covalently attached to the carrier, and the therapeutic compound. In one preferred embodiment, the carrier is a liposome having a surface coating of hydrophilic polymer chains where a folate ligand is attached to the free distal end of at least a portion of the hydrophilic polymer chains, and the therapeutic agent is entrapped in the liposomes. The composition is effective to achieve accumulation of the therapeutic compound in the cell in an amount sufficient to be cytotoxic. Also described are methods for administering a therapeutic compound to a person suffering from a multi-drug resistant condition.
Composition permettant d'administrer un composé thérapeutique à une cellule résistant à plusieurs médicaments chez une personne atteinte d'un cancer résistant aux médicaments. La composition est formée d'une molécule vecteur et d'un ligand de ciblage folate, qui est lié par covalence au vecteur, et du composé thérapeutique. Dans une forme de réalisation préférée, le vecteur est un liposome comportant un revêtement de surface de chaînes polymères hydrophiles, dans lequel un ligand folate est attaché à l'extrémité distale libre d'au moins une partie des chaînes polymères hydrophiles, et l'agent thérapeutique est piégé dans les liposomes. Cette composition est efficace pour assurer l'accumulation du composé thérapeutique dans la cellule dans une quantité suffisante pour être cytotoxique. On décrit également des procédés d'administration d'un composé thérapeutique à une personne présentant une résistance à plusieurs médicaments.
