The present invention relates to a rapidly disintegratable tablet for administration with or without the use of water. The tablet comprises at least one active substance and a mixture of excipients. The excipients provide desired characteristics and physical properties and when the tablet is sintered, excellent tablet binding characteristics are obtained. The tablet is intended for oral administration and dissolves in the presence of water. Also disclosed is a process for the preparation of a rapidly disintegratable tablet for administration with or without the use of water. The process comprising dissolving at least one carbohydrate and at least one structuring protein or polymer in a suitable solvent, wherein the solvent provides high porosity upon drying; spray-drying said dissolved mixture to obtain matrix or bead; dry blending at least one binding polymer, and at least one active drug with the matrix or bead to obtain a pretableting formulation or adding at least one active drug to the matrix or bead dissolved or dispersed components before spray-drying; compressing the formulation to obtain a tablet; and sintering the tablet for a sufficient time and temperature to allow the binding polymer to change status or melt and allow the polymer to resolidify as the temperature is reduced to ambient temperature.
La présente invention concerne un comprimé à désintégration rapide qu'on administre avec ou sans usage d'eau. Le comprimé comprend au moins une substance active et un mélange d'excipients. Les excipients présentent des caractéristiques et des propriétés physiques désirées, et le comprimé présente d'excellentes caractéristiques de liant lorsqu'il est fritté. Le comprimé s'admistre par voie orale et se dissout avec de l'eau. L'invention concerne également un procédé de préparation d'un comprimé à désintégration rapide qu'on administre avec ou sans usage d'eau. Le procédé consiste à dissoudre au moins un glucide et au moins une protéine ou un polymère structurant dans un solvant approprié, lequel solvant offre une porosité élevée après séchage. Le procédé consiste ensuite à sécher par pulvérisation ce mélange dissout pour obtenir une matrice ou perle; à mélanger à sec au moins un polymère cohésif et au moins un médicament actif avec la matrice ou perle pour obtenir une formule précurseur du comprimé, ou à ajouter au moins un médicament actif aux constituants dissouts ou dispersés de la matrice ou perle avant séchage par pulvérisation. Le procédé consiste enfin à comprimer la formule pour obtenir le comprimé; puis à fritter le comprimé pendant une durée et à une température suffisantes pour permettre au polymère cohésif de changer d'état ou de fondre dans un premier temps, puis de se resolidifier à mesure que la température baisse au niveau de la température ambiante.
