Methods for enhancing the production of interferons in animal cell culture are described. These methods rely on the manipulation of the cellular levels of certain inducers of interferon production, in particular cellular levels of double-stranded-RNA-dependent kinase (dsRNA-PKR, or PKR). In cell cultures that overproduce PKR, interferon synthesis is induced to high levels, and significant amount of interferon can be recovered without conventional induction of interferon by virus.
L'invention porte sur un procédé pour stimuler la production d'interférons dans une culture cellulaire animale. Ce procédé consiste à manipuler les niveaux cellulaires de certains inducteurs de production d'interférons, en particulier les niveaux cellulaires de kinase dépendante de RNA bicaténaire (dsRNA-PKR, ou PKR). Dans les cultures cellulaires qui produisent trop de kinase, la synthèse des interférons est induite à des niveaux élevés, et des quantités importantes d'interférons peuvent être récupérées sans induction classique des interférons par un virus.
