The invention relates to T cells transiently transfected with RNA, especially RNA encoding a T cell receptor and/or FoxP3, and to methods of transfecting T cells with RNA by electroporation. Compositions of the invention include an effector T cell transiently transfected with RNA encoding a T cell receptor (TCR) specific for an antigen, wherein the T cell demonstrates effector function specific for cells presenting the antigen in complex with an MHC molecule. Treg cells comprising an exogenous RNA encoding FoxP3 are also provided. The transfected T cells are useful for immunotherapy, particularly in the treatment of tumors, pathogen infection, autoimmune disease, transplant rejection and graft versus host disease.

L~invention concerne des lymphocytes T transitoirement transfectés avec de l~ARN, plus particulièrement de l~ARN encodant un récepteur de lymphocytes T et/ou le facteur de transcription FoxP3, et des procédés de transfection des lymphocytes T avec de l~ARN par électroporation. Des compositions objets de l~invention comprennent un lymphocyte T effecteur transitoirement transfecté avec de l~ARN encodant un récepteur de lymphocytes T (TCR) spécifique d~un antigène, ledit lymphocyte T affichant une fonction effectrice spécifique de cellules présentant l~antigène en complexe avec une molécule MHC. L~invention a également pour objet des lymphocytes Treg comprenant un ARN exogène encodant FoxP3. Les lymphocytes T transfectés sont utiles en immunothérapie, en particulier pour le traitement de tumeurs, d~infections pathogènes, de maladies auto-immunes, de rejets de transplantations et de réactions de greffes contre hôtes.
