The present invention is directed to a method of predicting a response to a chemotherapeutic regimen based on loss of heterozygosity at the thymidylate synthase locus in cancer tissue. This method comprises determining normal tissue genotype for thymidylate synthase of a patient; determining tumor tissue genotype for thymidylate synthase of said patient; comparing the normal tissue genotype with the tumor tissue genotype; determining whether a loss of heterozygosity at the thymidylate synthase locus has occurred in the tumor tissue based on the comparison of the genotypes; and predicting a response to the chemotherapeutic regimen based on the loss of heterozygosity in the tumor sample.
L'invention porte sur une méthode de prédiction de la réponse à un traitement probatoire de chimiothérapie en fonction en de la baisse d'hétérozygosité au niveau des locus de la thymidylate synthase de tissus cancéreux. Ladite méthode consiste: à déterminer le génotype de la thymidylate synthase d'un tissu normal du patient; à déterminer le génotype de la thymidylate synthase d'un tissu tumoral du patient; à comparer les deux génotypes; à déterminer s'il y a une baisse de l'hétérozygosité détectée au niveau des locus de la thymidylate synthase à l'aide de la comparaison; et à prédire la réponse à un traitement de chimiothérapie en fonction de la perte d'hétérozygosité de l'échantillon tumoral.
