Targeted cytotoxic T lymphocytes have a TcR comprising heterologous .alpha. and .beta. polypeptides, which heterologous polypeptides confer on the lymphocyte MHC Class I-restricted specificity for disease-causing target cells. The lymphocytes may be monovalent, having a single species of TcR conferring specificity for a single class of target cells. The lymphocytes find application as vaccines and in adoptive immunotherapy, e.g. cancer, AIDS or antiviral therapy.

Selon la présente invention, des lymphocytes T cytotoxiques ciblés comportent un TcR comprenant des polypeptides .alpha. et .beta. hétérologues, lesquels confèrent au lymphocyte une spécificité réduite en Classe I du système majeur d'histocompatibilité (MHC) pour les cellules cibles pathogènes. Ces lymphocytes peuvent être monovalents, et ne présenter qu'une seule espèce de TcR conférant la spécificité à une seule classe de cellules cibles. Ces lymphocytes trouvent une application comme vaccins et en immunothérapie adoptive, par exemple, dans la thérapie contre le cancer, le SIDA, ou antivirale.
