The present invention relates to transgenic non-human animals that are engineered to contain human immunoglobulin gene loci. In particular, animals in accordance with the invention possess human Ig loci that include plural variable (VH and V) gene regions. Advantageously, the inclusion of plural variable region genes enhances the specificity and diversity of human antibodies produced by the animal. Further, the inclusion of such regions enhances and reconstitutes B-cell development to the animals, such that the animals possess abundant mature B-cells secreting extremely high affinity antibodies.
La présente invention se rapporte à des animaux transgéniques non humains que l'on manipule par génie génétique de manière à y incorporer des loci de gènes d'immunoglobuline d'origine humaine. Les mammifères de cette invention possèdent notamment des loci d'immunoglobuline d'origine humaine qui incorporent plusieurs régions géniques variables (V¿H? et V?). Cette incorporation de plusieurs régions géniques variables permet avantageusement de renforcer la spécificité et la diversité d'anticorps humains produits par ledit mammifère. L'incorporation de telles régions permet en outre de renforcer et de reconstituer le développement de cellules bêta chez ces mammifères, de sorte que ces derniers possèdent des cellules bêta matures en abondance sécrétant des anticorps dotés d'une affinité extrêmement élevée.
