The present invention is directed to methods of detecting the presence of a bipolar mood disorder susceptibility locus in an individual, comprising analyzing a sample of DNA for the presence of a DNA polymorphism on the short arm of chromosome 18 between the telomere and D18S481, wherein the DNA polymorphism is associated with a form of bipolar mood disorder. The invention for the first time provides strong evidence of a susceptibility gene for bipolar mood disorder that is located in the terminal 5 cM region of the short arm of chromosome 18. The disclosure describes the use of linkage analysis and genetic markers in this 5 cM region to fine map the region and the use of genetic markers to genetically diagnose (genotype) bipolar mood disorder in individuals, to confirm phenotypic diagnoses of bipolar mood disorder, to determine appropriate treatments for patients with particular genotypic subtypes. Isolated polynucleotides useful for genetic linkage analysis of BP-I and methods for obtaining such isolated polynucleotides are also described.
Procédés de détection de la présence d'un locus de susceptibilité à la psychose maniaco-dépressive chez un sujet, qui consiste à analyser un échantillon d'ADN en vue d'y rechercher la présence d'un polymorphisme d'ADN sur le bras court du chromosome 18 entre le télomère et D18S481, ledit polymorphisme d'ADN étant associé à une forme de psychose maniaco-dépressive. La présente invention fournit pour la première fois des preuves solides de l'existence d'un gène de susceptibilité à la psychose maniaco-dépressive qui est situé dans la région terminale 5 cM du bras court du chromosome 18. La présente invention concerne l'utilisation de l'analyse de liaison et de marqueurs génétiques dans cette région 5 cM de manière à cartographier précisément ladite région et l'utilisation de marqueurs génétiques pour diagnostiquer génétiquement (génotype) la psychose maniaco-dépressive chez des sujets, pour confirmer les diagnostics de phénotypes de la psychose maniaco-dépressive et pour déterminer les traitements appropriés aux patients présentant des sous-types particuliers de génotype. Des polynucléotides isolés utiles pour l'analyse de liaison génétique de BP-I et des procédés permettant d'obtenir ces polynucléotides isolés sont également décrits.
