Provided, among other things, are compositions and methods for making sustained-release microspheres, as well as a microsphere delivery system for the sustained release of an active agent. The microsphere delivery system comprises a homogeneous mixture of biodegradable polymer, active agent, and a so-called release-modifying agent (including a pH-stabilizing agent), and provides protected and sustained release of active agents from the microsphere delivery system. According to the invention, the microspheres preferably are produced by an oil-in-water emulsion method that involves the production of a homogeneous oil phase prepared by mixing active agent and a release-modifying agent, such as arginine, with biodegradable polymer, each dissolved in organic solvent. The homogeneous oil phase desirably is then dispersed in an aqueous phase containing an emulsifying agent, followed by solvent removal, to produce the microspheres in which the active agent and release-modifying agent are distributed homogeneously throughout the biodegradable polymer matrix.

Sont présentées, entre autres, des compositions et méthodes pour la fabrication de microsphères à libération continue ainsi qu~un système d~administration de microsphères pour la libération continue d~un agent actif. Le système d~administration de microsphères est composé d~un mélange homogène de polymère biodégradable, d~un agent actif et d~un agent de modification de la libération (y compris un agent de stabilisation du pH) et assure la libération sûre et continue d~agents actifs à partir du système d~administration de microsphères. Selon l~invention, les microsphères sont de préférence produites par une méthode d~émulsion huile dans eau qui implique la production d~une phase huileuse homogène préparée par le mélange de l~agent actif et d~un agent de modification de la libération, comme l~arginine, avec un polymère biodégradable, chacun dissous dans un solvant organique. La phase huileuse homogène est alors de préférence dispersée dans une phase aqueuse contenant un agent émulsifiant, suivie de l~enlèvement du solvant, pour produire les microsphères où l~agent actif et l~agent de modification de la libération sont répartis de manière homogène dans l~ensemble de la matrice polymère biodégradable.
