A novel method for the preparation of a highly densified and at least partly, preferably fully or almost fully hydrated ceramic for use in the preparation of a pharmaceutical composition notably for controlled-release of one or more therapeutically, prophylactically and/or diagnostically active substance. The method involves a concomitant step of hydrating and densifying a bioresorbable and hydratable ceramic such as calcium sulphate. The invention also relates to compositions comprising such a highly densified ceramic. The pharmaceutical composition is useful for targeted and controlled local prolonged release of active substances, e.g. anti-cancer agents, whereby the spectrum and severity of side effects are minimized due to an optimized local concentration-time profile.
La présente invention concerne une nouvelle méthode d'élaboration d'une céramique hautement densifiée et au moins partiellement, préférentiellement entièrement ou presque entièrement hydratée, ladite céramique pouvant être employée dans l'élaboration d'une composition pharmaceutique, notamment pour la libération contrôlée d'un ou de plusieurs principes actifs thérapeutiques, prophylactiques et/ou diagnostiques. La méthode implique une étape simultanée d'hydratation et de densification d'une céramique biorésorbable et hydratable telle que le sulfate de calcium. L'invention concerne également des compositions comprenant une telle céramique hautement densifiée. La composition pharmaceutique peut être employée dans la libération prolongée ciblée et contrôlée localement de principes actifs, par exemple des agents anticancéreux, le spectre et la gravité des effets secondaires étant minimisés du fait de l'optimisation du profil local de concentration en fonction du temps.
