An assay for detecting inhibitors of the enzyme myeloperoxidase said method comprising: reacting myeloperoxidase with hydrogen peroxide and a chloride source to generate hypochlorus acid in the presence of a potential inhibitor of said myeloperoxidase; reacting any formed hypochlorous acid with a suitable amine to form the corresponding chloramine; Optionally removing any unreacted hydrogen peroxide; reacting any formed chloramine with a suitable detector molecule in the presence of iodide to form oxidised detector molecule; determining the amount of formed oxidised detector molecule by measuring the absorbance or fluorescence at a suitable wavelength; Identifying as inhibitors of myeloperoxidase those compounds which cause the amount of formed oxidised detector molecule to be decreased. And for use in diagnostic tests for myeloperoxidase activity is disclosed.
La présente invention concerne un essai permettant la détection d'inhibiteurs de l'enzyme myélopéroxydase, ledit procédé comprenant les étapes suivantes: réaction de myélopéroxydase avec du péroxyde d'hydrogène et une source de chlorure pour produire de l'acide hypochloreux en la présence d'un inhibiteur de potentiel de ladite myélopéroxydase; réaction de toute forme d'acide hypochloreux avec une amine appropriée pour former la chloramine correspondante; éventuellement élimination de tout péroxyde d'hydrogène n'ayant pas réagi; réaction de toute forme de chloramine avec une molécule de détection appropriée en la présence de iodure pour former une molécule de détection oxydée; détermination de la quantité de molécule de détection oxydée formée par mesure de la capacité d'absorption ou de la fluorescence à une longueur d'onde appropriée; identification en tant d'inhibiteurs de myélopéroxydase des composés qui provoquent la diminution de la quantité de molécule de détection oxydée formée. Le procédé de l'invention peut être utilisé dans le cadre d'essais diagnostiques de l'activité de la myélopéroxydase.
