A method is described to construct genetically modified strains of steroid degrading micro-organisms wherein multiple genes involved in steroid nucleus degradation are inactivated, such as steroid dehydrogenase genes. Examples for such genes are kstD1 and kstD2: strains with multiple inactivated steroid degrading enzyme genes can be used in the accumulation of steroid intermediates with a high yield. A preferred accumulation product is 9.alpha.- hydroxy-4-androstene-3,17-dione.

L'invention concerne un procédé permettant de construire des souches génétiquement modifiées de micro-organismes décomposant les stéroïdes, dans lequel de multiples gènes impliqués dans la décomposition du noyau stéroïde sont inactivés, tels que les gènes de stéroïde déshydrogénase. Ces gènes sont par exemple kstD1 et kstD2. Les souches comportant de multiples gènes inactivés d'enzymes décomposant les stéroïdes peuvent être utilisées dans l'accumulation de produits intermédiaires de stéroïdes à haut rendement. Un produit d'accumulation préféré est la 9.alpha.-hydroxy-4-androstène-3,17-dione.
