Compositions and methods for enhancing paracellular permeability at an absorption site in a subject are disclosed. The method includes: (a) administering an effective amount of a phospholipase C inhibitor to a subject at a time in which enhanced paracellular permeability is desired; and (b) enhancing paracellular permaeability in the subject at the the absorption site through the administering of the effective amount of the phospholipase C inhibitor. The disclosed compositions and methods provide enhanced absorption of a hydrophilic drug in a subject.

L'invention concerne des compositions et des méthodes permettant d'améliorer la perméabilité paracellulaire dans un site d'absorption chez un sujet. Cette méthode consiste (a) à administrer une dose nécessaire d'un inhibiteur de la phospholipase C à un sujet au moment où l'on souhaite obtenir une perméabilité paracellulaire améliorée, et (b) à améliorer la perméabilité paracellulaire chez un sujet dans le site d'absorption par administration de ladite dose nécessaire dudit inhibiteur de la phospholipase C. Les compositions et les méthodes de la présente invention permettent d'améliorer l'absorption d'un médicament hydrophile chez un sujet.
