The specification describes a method of preparing prostaglandin synthetase from a natural source, incubating the synthetase with diperlinolenic acid and purifying the prostaglandin E1 produced. The prostaglandin synthetase is prepared by homogenising a natural source of the synthetase in a halide of an alkali metal such as potassium chloride and separating the homogenised mixture into a liquid phase and a solid phase. The pH of the liquid phase is adjusted to a level in a range from 5 to 5.5 to cause the prostaglandin synthetase to form as a solid phase. The prostaglandin synthetase is then separated from the liquid phase and used to synthesise prostaglandin E1.
L'invention décrit un procédé qui consiste à fabriquer de la prostaglandine synthétase à partir d'une source naturelle, à incuber la synthétase au moyen de l'acide diperlinoléique et à purifier la prostaglandine E¿1? ainsi produite. On prépare la prostaglandine synthétase par l'homogénéisation d'une source naturelle de synthétase dans un halogénure de métal alcalin tel qu'un chlorure de potassium et par la séparation du mélange homogénéisé en une phase solide et une phase liquide. Le pH de la phase liquide est compris entre 5 et 5,5, ce qui permet à la prostaglandine synthétase de se former en phase solide. La prostaglandine synthétase est ensuite séparée de la phase liquide et utilisée pour synthétiser de la prostaglandine E¿1?.
