A rapid assay for single nucleotide polymorphism (SNP) detection that utilizes electronic circuitry on silicon microchips is disclosed. The method provides accurate discrimination of amplified DNA samples following electronic assisted transport, concentration, and attachment of DNA to selected electrodes (test sites). The test sites make up organized arrays of samples that are distinguished by using internal controls of dual labeled reporters comprising wild-type and mismatched sequences to validate the SNP genotype. This method has been used to discriminate the complex quadra-allelic SNP of mannose binding protein.

L'invention concerne un test rapide pour la détection du polymorphisme nucléotidioque unique (PSN) faisant appel à des circuits électroniques sur des micropuces de silicium. Cette technique effectue une discrimination précise d'échantillons d'ADN amplifiés après le transport assisté électroniquement, de la teneur et de la fixation de l'ADN aux électrodes sélectionnés (sites de test). Ces sites forment des réseaux organisés d'échantillons qui se distinguent par l'utilisation de commandes internes de doubles rapporteurs étiquetés comprenant des séquences de type sauvage et sans correspondance pour valider le génotype PSN. Cette technique sert à discriminer le PSN quadra-allèle complexe de la protéine de liaison du mannose.
