The present invention relates to apoptotic bodies derived from human tumor cells or cell lines recovered from patient's tumor biopsy and induced to apoptotis, said apoptotic bodies having the following characteristics: they maintain plasma membrane integrity, they are vesicles above about 0,1 µm, they contain intact mitochondria and cleaved nuclear DNA originating from the tumor cells, they present unmasked tumor antigens on their membranes, they present specific tumor and MHC antigens from the patient. The invention also provides new monocytes derived cells, which can be used as anti-tumor vaccines after integration of apoptotic bodies. Apoptotic bodies are phagocytosed and processed by monocyte derived antigen presenting cells and potentiate the effective tumor antigenic presentation to the immune system.
L'invention concerne des corps apoptotiques derivés de cellules ou de lignées cellulaires tumorales humaines provenant d'une biopsie de tumeurs chez des patients et provoquant une apoptose, lesdits corps apoptotiques présentant les caractéristiques suivantes : ils maintiennent l'intégrité de la membrane plasmatique; ils constituent des vésicules supérieurs à environ 0,1 µm ; ils contiennent des mitochondries intactes et de l'ADN nucléaire clivé provenant des cellules tumorales; ils présentent des antigènes tumoraux non masqués sur leurs membranes; ils présentent des antigènes spécifiques tumoraux et MHC provenant du patient. L'invention permet également d'obtenir de nouvelles cellules dérivées de monocytes pouvant être utilisées comme vaccins anticancéreux après intégration des corps apoptotiques. Les corps apoptotiques sont phagocytés et traités par des cellules présentant des antigènes, dérivées de monocytes et potentialisent la présentation antigénique tumorale efficace pour le système immunitaire.
