The novel 4-oxocyclic ureas and the pharmaceutically acceptable salts and esters thereof of the present invention are useful as antiarrhythmic and antifibrillatory agents and have following general structure: (I), wherein (a) X is a saturated or unsaturated, 5-, 6-, or 7-membered heterocycle or carbocycle; (b) R is selected from the group consisting of covalent bond, nil, heteroatom, carbonyl, heterocyclic ring, carbocyclic ring, alkyl, alkenyl, alkoxy, alkylamino, arylalkyl, aryloxy, aryl, acyloxy, and acylamino; (c) Y is a substituted or unsubstituted, saturated or unsaturated, 5-, 6-, or 7- membered heterocyclic ring or carbocyclic ring, or is nil; and wherein when R is nil, X and Y are fused ring systems; and when R is a covalent bond, X and Y are ring systems linked through a covalent bond; and when Y is nil, R is a covalent bond, and X is bound to L through R; (d) R1, R2, and R3 are independently selected from the group consisting of nil, Cl, F, Br, NH2, CF3, OH, S03H, CH3SO2NH, COOH, alkoxy; alkyl, alkoxycarbonyl, hydroxyalkyl, carboxyalkyl, aminoalkyl, acylamino and acyloxy; (e) L is selected from the group consisting of alkylamino, alkenylamino, alkylimino, alkenylimino, and acylamino; wherein the nitrogen atom thereof is bound to the nitrogen atom at the 1-position of the 4-oxocyclic urea moiety; (f) R4 is selected from the group consisting of alkyl, alkenyl, alkynyl, alkylacyl, and heteroalkyl; (g) A is a substituted or unsubstituted, saturated or unsaturated, straight-chain or branched, C1-C8 heteroalkyl; or a substituted or unsubstituted, saturated or unsaturated heterocycle having 5-, 6-, or 7-members; and A has one nitrogen atom, which is adjacent to R4; and (h) R5 is a substituted or unsubstituted C1 or C2 alkyl.
Les nouvelles urées cycliques de la présente invention sont utilisées comme agents anti-arythmiques et antifibrillants, et ont la structure générale (I) dans laquelle (a) X est un hétérocycle ou carbocycle saturé ou insaturé, à 5, 6 ou 7 éléments; (b) R es sélectionné dans le groupe composé par une liaison covalente, zéro, hétéroatome, carbonyle, cycle hétérocyclique, cycle carbocyclique, alkyle, alcényle, alcoxy, alkylamino, arylalkyle, aryloxy, acyle, acyloxy et acylamino; (c) Y est un cycle hétérocyclique ou carbocyclique à 5, 6 ou 7 éléments, substitué ou non substitué, saturé ou insaturé, ou est nul; et dans laquelle si R est nul, X et Y sont des systèmes de cycles fusionnés; et si R est une liaison covalente, X et Y sont des systèmes de cycles liés par une liaison covalente et X est lié à L par R, (d) R1, R2 et R3 sont sélectionnés indépendamment dans le groupe composé par zéro, Cl, F, Br, NH2, CF3, OH, SO3H, CH3SO2NH, COOH, alcoxy, alkyle, alcoxycarbonyle, hydroxyalkyle, carboxyalkyle, aminoalkyle, acylamino, et acyloxy; (e) L est sélectionné dans le groupe composé par alkylamino, alcénylamino, alkylamino, alcénylimino, et acylamino, dans lequel l'atome d'azote de celui-ci est lié à l'atome d'azote, en position 1, de la fraction cyclique de l'urée; (f) R4 est sélectionné dans le groupe composé par alkyle, alcényle, alkynyle, alkylacyle et hétéroalkyle; (g) A est un hétéroalkyle C1-C8 substitué ou non substitué, saturé ou insaturé, à chaîne droite ou ramifiée, ou un hétérocycle substitué ou non substitué saturé ou insaturé possédant 5, 6, ou 7 éléments; et comporte un atome d'azote adjacent à R4 et (h) R5 est un alkyle C1 ou C2 substitué ou non substitué; et par leurs esters et sels pharmaceutiquement acceptables.
